Maybe case-sensitive problem.
Try change "image": "/ISO/ubuntu-21.04-desktop-amd64.iso"  to  "image": "/iso/ubuntu-21.04-desktop-amd64.iso",

Recommend to use VentoyPlugson to automatically generate ventoy.json to avoid problem.

I copied the example on https://www.ventoy.net/en/plugin_persistence.html where iso is in capitals.
To prevent users going wrong it is better to change the example all lowercase.

It depends on the actual directory name. In my Ventoy USB I put all the ISO files in ISO directory. So the example is right for me.

You just add the persistence file and then add an entry into the ventoy.json file

That's what I've done and tested.

I want to use kali persistence.
I wrote ventoy.json like you, but there are no meau like "boot with persistence" or "boot without persistence" when I click kali.It will enter the selection menu of kali.I dont konw why.
